Chk2 and p53
WebCHK2 can phosphorylate p53 at Thr 18 or Ser 20 ( Shieh et al ., 2000 ). Phosphorylation at the Thr 18 site attenuates MDM2 (mouse double minute 2) binding to p53 ( Craig et al ., 1999; Schon et al ., 2002) and relieves p53 from negative control by MDM2. WebOur results indicate that MNK/Chk2-dependent modification of Drosophila p53 activates a global transcriptional response to DNA damage that induces error-prone DNA repair as well as intrinsic and extrinsic apoptosis pathways. WebE4F1 is essential for early embryonic mouse development and for controlling the balance between proliferation and survival of actively dividing cells. We previously reported that E4F1 is essential for the survival of murine p53-deficient cancer cells by controlling the expression of genes involved in mitochondria functions and metabolism, and in cell-cycle … WebMay 4, 2006 · Chk2 inactivation in humans with normal p53 leads to Li-Fraumeni syndrome , supporting a genetic connection between Chk2 and p53 and the existence of a Chk2-p53 axis [see the molecular interaction map (MIM) Fig. 1]. Families with Chk2 deficiencies have been identified among patients with increased risk of breast cancer .
WebJan 4, 2024 · by ATM and CHK2 activation. This initiates the induction of p53 expression and activation followed by TIP60-mediated p53 acetylation that triggers induction of PUMA and Bax, while DNA damage can also directly lead to PUMA induction. Once activated, PUMA selectively triggers the onset of mitochondrial-mediated WebMay 7, 2024 · The MRN complex and the acetyltransferase Tip60 activate ATM, which relays the damage signal to targets such as γH2AX, Chk2, p53, and Breast Cancer Susceptibility Gene 1 (BRCA1). γH2AX nucleates the site of damage, leading to the recruitment of several E3 Ubiquitin ligases that bring homologous repair factors (HR) …
WebDec 24, 2024 · The phosphorylation of a variety of substrates, including casein kinase (CK1), checkpoint kinase 1 (Chk2), and p53, regulates cancer cell viability by modulating many critical biochemical pathways that lead to cell cycle arrest, DNA repair, senescence, and death Full size image WebFeb 1, 2011 · The tumor suppressor p53 has been reported to be another key target of Chk2 in response to DNA damage. In fact, studies in knockout mice showed that Chk2 phosphorylates p53 on serine-20, and that this phosphorylation disrupts the p53-MDM2 interaction leading to the stabilization and accumulation of p53 after DNA damage ( 10, …

WebMay 4, 2006 · Our data revealed that ATM–Chk2–p53 DNA damage response (DDR) is activated in the absence of Brca1 during embryonic development and that DDR acts as a natural barrier to eliminate cells carrying DNA damage.
